New cyclic sulfamate compounds show potent activity against tuberculosis (TB) by inhibiting the essential KasA enzyme. This discovery offers a novel mechanism of action to combat drug-resistant Mycobacterium tuberculosis strains.
Area of Science:
- Microbiology
- Medicinal Chemistry
- Drug Discovery
Background:
- Tuberculosis (TB) remains the deadliest infectious disease globally.
- Current TB treatments are lengthy, have side effects, and face emerging drug resistance.
- Existing drugs target a limited number of bacterial pathways.
Purpose of the Study:
- To identify novel antimycobacterial agents with new mechanisms of action.
- To address the urgent need for effective treatments against multidrug-resistant TB.
- To explore cyclic sulfamate (CS) compounds as potential TB therapeutics.
Main Methods:
- Screening of cyclic sulfamate (CS) compounds for antimycobacterial activity.
- Whole genome sequencing of resistant mutants.
- Cell-wall damage reporter assays.
- Drug-target interaction modeling.
- Cell wall lipid profiling.
Main Results:
- Identified novel CS chemotypes with potent and selective activity against Mycobacterium tuberculosis (Mtb).
- Evidence strongly supports KasA inhibition as the mechanism of action for CS compounds.
- CS-resistant mutants retained susceptibility to first-line TB drugs, indicating a novel mechanism.
Conclusions:
- CS compounds represent a promising new class of antimycobacterial agents targeting the underexploited KasA enzyme.
- The study provides a foundation for medicinal chemistry optimization of CS compounds.
- This novel chemotype could lead to alternative treatment options for TB.
