Validation of the Microlab Listeria monocytogenes for Detection of Listeria monocytogenes in Select Foods: AOAC

Background:
Microlab Listeria monocytogenes is an all-in-one detection method that integrates, in a single disposable device, all elements to perform the analysis; a ready-to-use enrichment broth, a lateral flow immunochromatographic test, and a chemical agent for bacterial inactivation. It can be easily used on-site in food processing facilities.
Objective:
This report presents the validation data supporting AOAC Performance Tested Method SM 062502 certification of the Microlab Listeria monocytogenes test for Cheddar cheese, ice cream, smoked salmon, deli ham, and deli turkey. Environmental surfaces are not included in the validation study.
Methods:
The Microlab Listeria monocytogenes was compared to the ISO Standard 11290:2017 method for the detection of L. monocytogenes in select foods. The comparison study used 30 unpaired test portions for all matrixes. Inclusivity and exclusivity study evaluated 50 L. monocytogenes and 36 non-L. monocytogenes isolates. The product consistency/stability study evaluated three separate lots in an accelerated shelf-life design. Robustness was evaluated by modifying three operational parameters.
Results:
Inclusivity and exclusivity testing showed that the Microlab Listeria monocytogenes method detects L. monocytogenes strains while excluding the non-Listeria monocytogenes strains. L. innocua or L. marthii may yield positive results in foods at levels above 87 CFU/25 g. In the matrix studies, no statistically significant differences were observed between the candidate and reference methods. Minor procedural variations (enrichment time, time to read results, and enrichment temperature) did not affect assay performance. No significant differences were found between recently manufactured lots and those near expiration.
Conclusions:
The reported data confirm that Microlab Listeria monocytogenes reliably detects L. monocytogenes in the tested food matrixes. The method is based on presence/absence detection, with a limit of detection of 1 CFU per test portion.
