Abstract:
Rehmannia glutinosa, a widely used traditional Chinese medicinal herb and dietary therapeutic, is rich in polysaccharides. In this experiment, column chromatography was employed to separate and purify the polysaccharides from Rehmannia glutinosa, yielding the homogeneous polysaccharide RGP70-2-1. Structural analysis of RGP70-2-1 was conducted using various methods, including HPLC, FT-IR, GC-MS, NMR, and SEM. The results indicate that RGP70-2-1 is a high-molecular-weight structure composed of five monosaccharides (mannose, rhamnose, galactose, arabinose, and glucose), with 14 types of linkages. Building upon this structural framework, this study employed molecular docking and in vitro assays to demonstrate that RGP70-2-1 significantly inhibits α-glucosidase and α-amylase activities, indicating its potential hypoglycemic effect. The work enriches the primary structural information on Rehmannia glutinosa polysaccharides and lays a foundation for future investigations into their bioactivities and structure-function relationships.
