Guided discovery of anti-hyperuricemic active molecules from Dioscorea septemloba by activity labelled molecular

Abstract:
Six new molecules (1-6) were obtained from the rhizomes of Dioscorea septemloba according to the ALMN workflow showed anti-hyperuricemic activity. The constructed ALMN could clearly visualize the activities and the structure nodes, which can facilitate the annotation of the active candidates. As for the new compounds, 1 and 2 were identified as steroid saponins, 3 as pentacyclic triterpene saponins, 4 and 5 as diarylheptanes, and 6 as a dihydrophenanthrene polycyclic aromatic hydrocarbon. Structures of these new compounds were determined by NMR analysis and ECD calculation. All the obtained new compounds were assayed for their anti-hyperuricemic activities using the HK-2 cell model induced by adenosine combined with XOD, all the six compounds showed significant anti-hyperuricemic effects, which were consistent with the obtained activity annotations by ALMN.
