    Summary

    Low-intensity focused ultrasound (LIFU) effectively reduced tic-like behaviors and brain activity in a Tourette syndrome mouse model. This noninvasive neuromodulation shows promise for future human tic disorder treatments.

    Background:

    • Tourette syndrome (TS) treatment often relies on pharmacological therapies with limited efficacy and side effects.
    • Current neuromodulatory therapies like DBS and TMS have limitations in invasiveness and spatial resolution.
    • Low-intensity focused ultrasound (LIFU) offers a noninvasive alternative for neurological disorders.

    Purpose of the Study:

    • To investigate the efficacy of LIFU as a novel therapeutic approach for tic disorders.
    • To establish effective LIFU stimulation parameters for alleviating tic-like movements.
    • To assess LIFU's potential for treating Tourette syndrome.

    Main Methods:

    • Development of a bicuculline-induced mouse model of tic disorder.
    • Validation of tic-like features using electrophysiological, neural activity, and behavioral analyses.
    • Application of clinically relevant LIFU stimulation (250-kHz) targeting the secondary motor cortex (M2).

    Main Results:

    • LIFU stimulation significantly reduced the frequency of tic-like movements by 51%.
    • Pathological electrophysiological spikes in the M2 were reduced by 87% following LIFU.
    • Demonstrated alleviation of tic symptoms in a preclinical model.

    • LIFU brain stimulation is effective in alleviating tic symptoms in a preclinical model of Tourette syndrome.
    • The established stimulation parameters are clinically compatible, supporting translational research.
    • LIFU presents a promising noninvasive therapeutic strategy for tic disorder treatment.
