Photocatalyzed Nitrene Insertion into C-O Bonds: Access to Benzisoxazoline-3-ones

Abstract:
The first photoinduced formal nitrene insertion into C-O bonds is established with sulfilimines as free nitrene precursors, providing facile access to N-substituted benzisoxazoline-3-ones under mild and additive-free conditions. DFT calculations indicate a mechanism proceeding via a singlet nitrene intermediate. Notably, the products demonstrate potent antitumor activity against a range of human cancer cell lines, highlighting their promise for anticancer drug discovery.
