Basal cytoplasmic calcium levels regulate the proliferation rate of Caenorhabditis elegans germ cells. The TRPM7-related GON-2 calcium channel is essential for maintaining these calcium levels and subsequent cell division.
Area of Science:
- Cell Biology
- Developmental Biology
- Molecular Biology
Background:
- The Caenorhabditis elegans germline is a key model for studying germ cell biology.
- Calcium signaling plays a role in various cellular processes, but its role in regulating germline proliferation is not fully understood.
Purpose of the Study:
- To investigate the relationship between cytoplasmic calcium levels and germ cell proliferation in C. elegans.
- To identify the molecular mechanisms regulating basal calcium levels in the germline.
Main Methods:
- Development of an integrated ratiometric calcium reporter for comparative analysis of germ cell calcium.
- Genetic manipulation of the GON-2 calcium channel and other calcium transporters (plasma membrane calcium ATPase, SERCA).
- Observation of germ cell proliferation rates under various physiological and genetic conditions.
Main Results:
- A positive correlation was observed between the rate of germ cell proliferation and basal cytoplasmic calcium levels.
- Conditions decreasing proliferation (starvation, altered signaling) reduced calcium levels.
- Overproliferating tumor cells exhibited increased basal calcium levels.
- Inactivation of the GON-2 calcium channel significantly decreased both basal calcium levels and proliferation.
- Inhibition of calcium efflux (plasma membrane calcium ATPase) or uptake (SERCA) increased basal calcium and proliferation.
Conclusions:
- Basal cytoplasmic calcium levels act as a rheostat to modulate C. elegans germ cell proliferation.
- The GON-2 calcium channel is crucial for maintaining basal cytoplasmic calcium levels in the germline.
- This study identifies the germline as the first C. elegans tissue where proliferation rate is directly regulated by basal calcium levels.
