Proteomics

7.5K
A proteome is the entire set of proteins that a cell type produces. We can study proteomes using the knowledge of genomes because genes code for mRNAs, and the mRNAs encode proteins. Although mRNA analysis is a step in the right direction, not all mRNAs are translated into proteins.
Proteomics is the study of proteomes' function. It involves the large-scale systematic study of the proteome to denote the protein complement expressed by a genome. Scientist Mark Wilkins coined the term...

Summary

The novel 16plex isobaric tags (IBT-16plex) reagent shows promise for single-cell proteomics. This study optimized IBT-16plex labeling and demonstrated its comparability to TMTpro-16plex for profiling mouse spleen cells.

Area of Science:

  • Proteomics
  • Mass Spectrometry
  • Cell Biology

Background:

  • Isobaric tag-based mass spectrometry is established for single-cell proteomics.
  • 16plex isobaric tags (IBT-16plex) are a novel reagent not yet applied to single-cell analysis.

Purpose of the Study:

  • To evaluate the suitability of IBT-16plex for proteomic quantification at the single-cell level.
  • To optimize labeling conditions and assess performance compared to existing reagents.

Main Methods:

  • Systematic evaluation of IBT-16plex reaction conditions, including mass ratios and carrier protein strategies.
  • Parallel labeling of single-cell peptides from mouse spleen tissue using IBT-16plex and TMTpro-16plex.
  • Bioinformatic analysis for protein identification, quantification, and cell type clustering.

Main Results:

  • Optimal IBT-16plex labeling achieved with a mass ratio >10,000 and a 200-fold excess of carrier proteins.
  • IBT-16plex relative quantification demonstrated comparability to TMTpro-16plex.
  • Successful profiling and quantification of single cells from mouse spleen, clustering into monocytes, dendritic cells, macrophages, and T cells.

Conclusions:

  • IBT-16plex is a viable and effective reagent for single-cell proteomics.
  • This method enables proteomic analysis of single cells isolated directly from animal tissues.
