Abstract:
Adventitious root (AR) development is controlled by both endogenous and environmental factors, among which light is an important environmental factor to promote root development. However, knowledge about the mechanisms controlling light-mediated AR formation in etiolated hypocotyl cuttings is not fully understood. In this study, genetic and biochemical analyses showed that light activates the transcription of the photomorphogenic gene HY1 to promotes AR pre-branch sites initiation, and subsequently promotes AR development in de-rooted Arabidopsis hypocotyls. By tissue-specific complementation experiments, we showed that plastid-localized AtHY1 in pericycle cells was capable of activating AR formation. Moreover, transcriptome profiling and genetic evidence revealed that HY1 induces the expression of HY5 and its homolog HYH, which act as downstream regulator to control AR formation by promoting auxin transport and conjugation. Collectively, our results highlight the essential role of light during the induction phase of HY1-mediated AR formation in Arabidopsis hypocotyl cuttings.
