Abstract:
Seed size and weight are critical yield-determining traits in soybean, yet their underlying genetic mechanisms remain incompletely clarified. To address this, we developed a newly expanded recombinant inbred lines (RILs) population, designated H3G, derived from selected lines of the GB13 RIL population established in our previous study. Employing multiple quantitative trait loci (QTL) mapping models and cross-population comparative analyses, we identified nine core genomic regions associated with seed size and weight traits. Finally, through systematic candidate gene identification and functional validation, we confirmed GmUbiE3 as a novel negative regulator of seed size and weight. CRISPR/Cas9-mediated knockout of GmUbiE3 resulted in a significant 28.07 % increase in soybean hundred-seed weight. These findings provide fundamental insights into the genetic control of soybean seed size and weight and offer valuable targets for molecular breeding approaches aimed at soybean yield improvement.
