Lipids as Anchors01:32

Lipids as Anchors

In the plasma membrane, the lipids forming the bilayer can also act as an anchor to tether proteins to the membrane. The three main types of lipid anchors found in eukaryotes are – prenyl groups, fatty acyl groups, and glycosylphosphatidylinositol or GPI groups. Prenyl and fatty acyl groups act as anchors on the cytosolic surface of the membrane, whereas GPI anchors proteins on the extracellular side.
The carboxy-terminal of most of the prenylated proteins, such as Ras proteins, contains the...

Area of Science:

  • Materials Science
  • Biotechnology
  • Pharmacology

Background:

  • Curcumin (CUR) exhibits therapeutic potential but suffers from poor solubility, stability, and bioavailability.
  • Encapsulation is a key strategy to overcome curcumin's limitations for enhanced therapeutic applications.

Purpose of the Study:

  • To develop a novel microencapsulation method for curcumin using a bio-based phase change material (PCM).
  • To characterize the curcumin-loaded PCM microcapsules and evaluate their drug release profile.

Main Methods:

  • Microencapsulation of curcumin using a eutectic mixture of stearic and lauric acids as a bio-based PCM.
  • Characterization via FT-IR, SEM, DSC, and TGA.
  • In vitro release studies in PBS (pH 7.4) at 37°C and 45°C.

Main Results:

  • Successful microencapsulation of curcumin within the bio-based PCM was confirmed.
  • Curcumin-loaded microcapsules showed well-defined morphology.
  • Sustained curcumin release observed, with ~50% and ~60% released at 37°C and 45°C respectively over 24 hours.

Conclusions:

  • Bio-based PCMs are effective carriers for curcumin microencapsulation and controlled delivery.
  • This approach holds promise for improving curcumin's therapeutic efficacy in drug delivery systems.
