Bulbil formation is a complex biological process that requires coordinated regulation by functional proteins. The DOF gene family, a group of plant-specific transcription factors, plays crucial roles in meristem differentiation and branching across species. However, the transcriptional regulation of DOF genes during bulbil development remains poorly understood. In this study, we systematically identified DOF family genes in lily (L. lancifolium). Yeast two-hybrid (Y2H) analysis demonstrated that LlDOF3.6.3, a key regulator of bulbil formation in L. lancifolium, physically interacts with LlWOX11, a known positive regulator of bulbil formation. Furthermore, we discovered that LlWOX11 binds to a novel AGAG motif located in the promoter region of LlDOF1.7. Our results elucidate the molecular mechanism whereby LlWOX11 activates LlDOF1.7 expression through direct promoter binding. These findings significantly enhance our understanding of the regulatory network involving LlDOFs and LlWOX11 in bulbil formation.
