Tumor Progression02:07

Tumor Progression

7.2K
Tumor progression is a phenomenon where the pre-formed tumor acquires successive mutations to become clinically more aggressive and malignant. In the 1950s, Foulds first described the stepwise progression of cancer cells through successive stages.
Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...

Summary

The protein BAG3 (Bcl-2-associated athanogene 3) is highly expressed and secreted in many cancers, suggesting its role in tumor survival. Elevated BAG3 levels in patient serum may indicate liver, pancreatic, or ovarian cancer.

Area of Science:

  • Oncology
  • Molecular Biology
  • Biochemistry

Background:

  • The protein BAG3 (Bcl-2-associated athanogene 3) is recognized as a stress-induced factor involved in cell survival across various cancer types.
  • Its precise role and expression patterns in diverse malignancies require further elucidation.

Purpose of the Study:

  • To investigate the expression and secretion of BAG3 in a broad range of cancer cell lines and primary tumors.
  • To evaluate BAG3 as a potential diagnostic biomarker and therapeutic target in oncology.

Main Methods:

  • Analysis of BAG3 expression and secretion in 24 cancer cell lines from ten distinct cancer types.
  • Comparison of cellular BAG3 levels with serum BAG3 concentrations in cancer patients and healthy controls.
  • Immunohistochemical assessment of BAG3 expression in primary tumor samples, correlating with tumor grade.

Main Results:

  • BAG3 demonstrated ubiquitous expression and secretion in all analyzed cancer cell lines.
  • Serum BAG3 levels were significantly elevated in patients diagnosed with liver, pancreatic, and ovarian cancers compared to healthy individuals.
  • Widespread high BAG3 expression was confirmed in multiple tumor types, frequently associated with higher tumor grade.

Conclusions:

  • BAG3 is a key regulator of tumor cell survival, consistently expressed and secreted across various cancers.
  • Elevated serum BAG3 levels show potential as a biomarker for specific cancer types, including liver, pancreatic, and ovarian cancers.
  • BAG3 represents a promising therapeutic target for cancer treatment strategies.
