Brevibacillus nigriterrae sp. nov., a Novel Denitrification Bacterium Isolated from Black Soil

Abstract:
A Gram-stain-positive, rod-shaped, endospore-forming, immotile and aerobic bacterial strain, designated as SYSU BS000544T, was isolated from a black soil sample in Harbin, Heilongjiang province, China. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain SYSU BS000544T belongs to the genus Brevibacillus, and showed the highest sequence similarity to Brevibacillus ginsengisoli KCTC 13938T (98.09%). Strain SYSU BS000544T was found to grow at 25-37 °C (optimum, 30 °C) and pH 6.0-7.0 (optimum, pH 6.5) and in the presence of 0% (w/v) NaCl. Polar lipids comprised of phosphophatidylinositol, phosphatidylethanolamine, one unidentified aminophospholipid, and two unidentified polar lipids. The major cellular fatty acids (> 5%) were iso-C17:1 ω9c and/or 10-methyl C16:0, iso-C16:0, iso-C17:0, iso-C14:0 and anteiso-C15:0. The predominant respiratory quinone was MK-7. The genomic G + C content was 48.84% based on the whole genome analysis. The average nucleotide identity (≤ 78.88%) and digital DNA-DNA hybridization (≤ 15.7%) values between strain SYSU BS000544T and other members of the genus Brevibacillus were all lower than the threshold values recommended for distinguishing novel species. Based on the results of phenotypic, chemotaxonomic and phylogenetic analyses, strain SYSU BS000544T represents a novel species of the genus Brevibacillus, for which the name Brevibacillus nigriterrae sp. nov. is proposed. The type strain is SYSU BS000544T (= MCCC 1K09332T = KCTC 43752T).
