Researchers developed a 3D culture system to create reconstituted human testes (rhTestes). This platform supports human germ cells (hGCs) and aids in studying male gametogenesis and infertility.
Area of Science:
- Reproductive Biology
- Developmental Biology
- Cell Biology
Background:
- Human germ cells (hGCs) migrate to gonads early in development.
- Post-sex determination, gonadogenesis and gametogenesis proceed distinctly in males and females.
- Interactions between hGCs and Sertoli cells in male testes are poorly understood.
Purpose of the Study:
- To establish a 3D culture platform for human fetal testes (rhTestes).
- To investigate factors influencing rhTestes organization and hGC survival.
- To explore interactions between testicular cell types in vitro.
Main Methods:
- Dissociated second-trimester human fetal testes were used for reconstitution.
- The study examined the impact of stress inhibitors and cell ratios (Sertoli cells, interstitial cells) on rhTestes.
- DDX4+ hGCs were monitored for survival and location within the 3D cultures.
Main Results:
- A 3D culture system successfully reconstituted human testes (rhTestes).
- DDX4+ hGCs were observed for at least two weeks, located at the periphery of rhTestes.
- The study identified factors affecting rhTestes organization and hGC numbers.
Conclusions:
- This 3D culture platform supports human fetal testes reconstitution.
- The system facilitates the study of testicular cell interactions.
- This platform offers potential insights into human male gametogenesis and infertility.
