Cancer-Critical Genes II: Tumor Suppressor Genes

9.3K
Genes usually encode proteins necessary for the proper functioning of a healthy cell. Mutations can often cause changes to the gene expression pattern, thereby altering the phenotype.
When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors.
Such genes that act...

Area of Science:

  • Oncology
  • Immunology
  • Genomics

Background:

  • Ovarian cancers exhibit diverse immunogenomic profiles despite similar histology.
  • Optimal treatment requires understanding these immunogenomic differences.

Purpose of the Study:

  • To identify key drivers of inter-tumour heterogeneity in ovarian cancer.
  • To explore immunogenomic profiles beyond histological subtypes.

Main Methods:

  • Integrated analysis of somatic mutation, mRNA expression, and multiplex immunofluorescence data.
  • Utilized a multi-modal latent variable model on 197 ovarian cancer patients.

Main Results:

  • Gene expression and immune infiltration in the tumour core/margin drive most heterogeneity.
  • Somatic mutations differentiate high-grade serous from other subtypes.
  • Endometrioid ovarian cancers show distinct immunogenomic features with improved prognosis.

Conclusions:

  • Ovarian cancer classification and treatment should evolve beyond histology.
  • Patient-specific tumour microenvironments offer targeted immuno-oncology opportunities.
  • Collagen gene expression correlates with fibroblast infiltration and reduced proliferation.
