Tumor necrosis factor-alpha (TNF-α) impacts microRNA-4693-5p (miR-4693-5p) levels in rheumatoid arthritis (RA) cells. This microRNA influences key proteins and pathways, showing therapeutic potential for RA treatment.

Area of Science:

  • Biochemistry
  • Immunology
  • Molecular Biology

Background:

  • Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by symmetrical joint inflammation.
  • Dysregulated microRNAs (miRNAs) are implicated in RA pathogenesis.
  • Previous studies indicated decreased miR-4693-5p in RA patients' peripheral blood mononuclear cells (PBMCs), affecting apoptosis, inflammatory cytokines, and reactive oxygen species (ROS).

Purpose of the Study:

  • To investigate the regulatory effect of TNF-α on miR-4693-5p expression in RA.
  • To examine the downstream effects of miR-4693-5p on RA-associated proteins and pathways.
  • To evaluate the therapeutic potential of miR-4693-5p in RA.

Main Methods:

  • Quantitative real-time PCR (qRT-PCR) assessed TNF-α's impact on miR-4693-5p in SW982 cells.
  • SWATH-MS identified differentially expressed proteins in RA-fibroblast-like synoviocytes (RA-FLS) regulated by miR-4693-5p.
  • Bioinformatic analysis (miRNA target prediction, Cytoscape) and validation (Western blotting, Mitosox, MTT assay, Hoechst staining) were performed. The collagen-induced arthritis (CIA) rat model was used for in vivo validation.

Main Results:

  • TNF-α was found to regulate miR-4693-5p expression in RA-mimic SW982 cells.
  • SWATH-MS identified 49 significantly regulated proteins, including FN-1, YWHAZ, SOD2, and CALD1, directly targeted by miR-4693-5p.
  • miR-4693-5p in vitro regulated extracellular matrix (ECM) proteins, epithelial-to-mesenchymal transition (EMT), VEGF signaling, mitochondrial ROS, cell proliferation, and apoptosis. Protein expression was validated in the CIA rat model.

Conclusions:

  • miR-4693-5p plays a crucial role in regulating RA pathogenesis by influencing cell adhesion, cytoskeleton organization, angiogenesis, and cell death pathways.
  • These findings highlight miR-4693-5p as a promising therapeutic target for RA treatment and disease management.

The JAK-STAT Signaling Pathway01:20

The JAK-STAT Signaling Pathway

Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. The dimerization brings the cytosolic JAKs together that trans-phosphorylate and activates each other. The activated JAKs now phosphorylate cytosolic tails of the cytokine receptors, which serve as binding sites for adaptor proteins such as  SH2...

T Cell Types and Functions01:24

T Cell Types and Functions

When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. The differentiation of CD4 T cells into helper T cell subsets, such as Th1, Th2, and Th17 cells, is dependent on the antigen type, antigen-presenting cell, and regulatory cytokines.
Th1 cells stimulate dendritic cells to express necessary co-stimulatory molecules on their surfaces for...

Genome-wide Association Studies-GWAS

Genome-wide association studies or GWAS are used to identify whether common SNPs are associated with certain diseases. Suppose specific SNPs are more frequently observed in individuals with a particular disease than those without the disease. In that case, those SNPs are said to be associated with the disease. Chi-square analysis is performed to check the probability of the allele likely to be associated with the disease.
GWAS does not require the identification of the target gene involved in...

TGF - β Signaling Pathway01:16

TGF - β Signaling Pathway

The TGF-β signaling pathway regulates cell growth, differentiation, adhesion, motility, and development. TGF-β ligands that induce TGF-β signaling are synthesized in their latent form. Several proteases or cell surface receptors such as integrins act upon the latent form, releasing the active ligand. There are three types of mammalian TGF-βs: (TGF-β1, TGF-β2, and TGF-β3) that bind as homodimers or heterodimers to TGF-β receptors. The TGF-β receptors...

Inflammatory Response01:28

Inflammatory Response

An inflammatory response is a localized, nonspecific immune reaction that occurs when a tissue is injured. It is characterized by redness, swelling, heat, and pain, which are commonly called the cardinal signs and symptoms of inflammation. Inflammation can sometimes result in a loss of function.
Inflammation can be triggered by various stimuli, such as impact, abrasion, chemical irritation, infections, and extreme hot or cold temperatures. These can damage cells and connective tissue fibers,...

MicroRNAs01:22

MicroRNAs

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...
