Lorlatinib, an ALK inhibitor, shows promise for treating frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS) by rescuing synaptic deficits caused by C9orf72 gene mutations. This therapy normalizes neuronal function and enhances resilience to excitotoxicity.
