Background:

  • Multiple Systems Atrophy (MSA) is a rare, sporadic, age-related synucleinopathy causing Parkinson-like motor symptoms and ataxia.
  • Pathologically, MSA involves glial cytoplasmic inclusions (GCI) of alpha-synuclein (αSyn) in oligodendrocytes, leading to demyelination.
  • Currently, no definitive therapy exists for MSA, with treatment limited to symptom management.

Purpose of the Study:

  • To investigate the efficacy of systemically delivered αSyn antisense oligonucleotides (ASO) in a mouse model of MSA.
  • To determine if αSyn ASO reduces αSyn accumulation in oligodendrocytes and mitigates MSA-associated pathology.

Main Methods:

  • Utilized a peptide-mediated delivery system to administer αSyn ASO systemically in an MSA mouse model.
  • Administered monthly treatments of αSyn ASO and assessed myelination, oligodendrocyte numbers, gliosis, and αSyn levels.

Main Results:

  • Monthly αSyn ASO treatments increased myelination in the corpus callosum and cerebellum.
  • Observed increased oligodendrocyte numbers and reduced gliosis.
  • No significant changes in overall αSyn levels were detected; peptide-mediated delivery was not effectively taken up by oligodendrocytes.

  • Systemic αSyn ASO administration partially alleviated MSA pathology in a mouse model.
  • The lack of oligodendrocyte uptake by the current delivery method indicates a need for alternative or supplementary strategies for effective MSA treatment.
