Targeted Cancer Therapies

The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. Summary

Claudin-18.2 (CLDN18.2) is highly expressed in specific biliary tract cancer (BTC) subtypes, while Trop-2 is broadly expressed. These findings support CLDN18.2- and Trop-2-targeted therapies for BTC patients.

Background:

  • Biliary tract cancers (BTCs) are aggressive with limited treatment options.
  • Claudin-18.2 (CLDN18.2) and Trophoblast cell surface antigen 2 (Trop-2) are promising targets in other epithelial cancers.
  • CLDN18.2 and Trop-2 expression in BTC is not well understood.

Purpose of the Study:

  • To evaluate CLDN18 and Trop-2 expression in BTC.
  • To assess their potential as therapeutic targets for BTC.

Main Methods:

  • Retrospective analysis of 636 BTC patients.
  • Immunohistochemistry used to assess CLDN18 and Trop-2 expression.
  • Statistical analysis of expression and clinicopathologic features.

Main Results:

  • CLDN18 was positive in 5.2% of BTC patients, notably in perihilar cholangiocarcinoma (pCCA).
  • Trop-2 was highly expressed in 69.5% of patients, with low expression in intrahepatic cholangiocarcinoma (iCCA).
  • Neither biomarker correlated significantly with overall or disease-free survival.

Conclusions:

  • CLDN18 shows specific expression in BTC subtypes like pCCA and gallbladder cancer.
  • Trop-2 demonstrates broad expression across BTC.
  • Findings support CLDN18.2- and Trop-2-targeted therapies for BTC.
