Genome-wide Association Studies-GWAS

15.2K
Genome-wide association studies or GWAS are used to identify whether common SNPs are associated with certain diseases. Suppose specific SNPs are more frequently observed in individuals with a particular disease than those without the disease. In that case, those SNPs are said to be associated with the disease. Chi-square analysis is performed to check the probability of the allele likely to be associated with the disease.
GWAS does not require the identification of the target gene involved in...

iPS Cell Differentiation

3.0K
The ability of induced pluripotent stem cells or iPSCs to differentiate into most body cell types has stimulated repair and regenerative medicine research over the past few decades. iPSC-derived blood cells, hepatocytes, beta islet cells, cardiomyocytes, neurons, and other cell types can repair injuries or regenerate damaged tissue in diseases such as diabetes and neurodegenerative disorders.

Area of Science:

  • Ophthalmology
  • Dermatology
  • Genetics

Background:

  • Psoriasis and age-related macular degeneration (AMD) share potential immune-related mechanisms.
  • Limited research has explored the connection between psoriasis and AMD.
  • This study investigates a potential causal relationship in European cohorts.

Purpose of the Study:

  • To assess the causal link between psoriasis and age-related macular degeneration (AMD).
  • To explore the role of immune-related pathophysiologic characteristics in both conditions.

Main Methods:

  • Utilized single-nucleotide polymorphisms (SNPs) for psoriasis as instrumental variables (IVs) from the FinnGen genome-wide association study (GWAS).
  • Obtained AMD GWAS data from the International AMD Genomics Consortium.
  • Employed a two-sample Mendelian randomization (MR) study with inverse-variance weighted, weighted median, and MR-Egger methods, alongside MR-Pleiotropy Residual Sum and Outlier (MR-PRESSO) testing.

Main Results:

  • Observed significant causal associations between psoriasis and AMD.
  • The weighted median method indicated an odds ratio (OR) of 1.09 (95% CI: 1.03-1.16, p=0.005).
  • The MR-PRESSO test yielded an OR of 1.04 (95% CI: 1.00-1.09, p=0.043).

Conclusions:

  • A potential causal association between psoriasis and AMD was identified.
  • Inflammation should be further investigated as a risk factor for AMD.
  • These findings may inform future research on shared etiological pathways.
