Purpose of the Study:

  • To survey raspberry viruses and their associated arthropods in the Czech Republic.
  • To identify prevalent viral pathogens and potential new hosts or vectors.
  • To assess viral genetic variability for improved disease management.

Main Methods:

  • RT-(q)PCR was used to test 257 plant and 151 arthropod samples for 14 viruses.
  • Sequencing of PCR amplicons was performed on 92 viral isolates.
  • Experimental transmission studies were conducted to identify vectors.

Main Results:

  • Raspberry bushy dwarf virus (RBDV, 51.8%) was the most common, followed by black raspberry necrosis virus (BRNV, 42.0%) and raspberry leaf mottle virus (RLMV, 28.4%).
  • RBDV was detected in a new host, *Sambucus nigra*. Mixed infections of RLBV and a novel virus (RaVA) were found in wild *Rubus occidentalis*.
  • Seven of 39 arthropod species carried viruses; *Amphorophora rubi idaei* and *Aphis idaei* were confirmed vectors. High viral genetic variability was observed.

Conclusions:

  • This survey provides crucial data on raspberry virus prevalence and diversity in the Czech Republic.
  • New host and vector associations were identified, necessitating updated management approaches.
  • Continued monitoring and research are essential to mitigate virus threats to raspberry cultivation.
