The screening of strains for organic acid biosynthesis is a key step to enhance its production. In view of the problems such as long-time consumption, complex instruments, large sample dosage, and low screening efficiency existing in traditional strain screening methods, this study had developed a high-throughput microdroplet screening system based on surface-enhanced Raman spectroscopy (SERS). This platform achieved in-situ analysis of bacterial metabolites within individual microdroplets by integrating microfluidic chips, SERS, and laser detection modules. High level succinic acid (SA), d-phenyllactic acid (d-PLA), and acrylic acid (AA)-producing engineered strains were obtained using our system. Particularly, 138.92 g/L SA was produced. Meanwhile, using bamboo hydrolysate instead of glucose as the fermentation substrate, 70.31 g/L SA was produced. This study verified the great potential of the system in selecting strains for organic acid production, which is expected to be widely used in green fabrication and other fields.
