A Tailing Method of Quality Control on MALBAC-Based Whole-Genome Amplification Products

Abstract:
Single-cell whole-genome sequencing (scWGS) has illuminated new perspectives on genetic diversity across different cellular conditions. Despite this, the amplification of DNA from sparsely populated formalin-fixed tissues during the whole-genome amplification (WGA) process remains a formidable challenge. We introduce an enhanced quality control protocol for WGA products generated using the multiple annealing and looping-based amplification cycles (MALBAC) technique. This approach presents a cost-efficient, high-throughput quality control method, delivering a precise assessment of WGA efficacy with 100% accuracy on a test set of 128 samples.
