Abstract:
T cells are key players of the cellular immune system, able to detect and fight pathogens' invasion. T cells recognize pathogenic peptides presented by proteins called Human Leukocyte Antigens (HLA) in humans. The molecular mechanism of T cells' interaction with peptide-HLA (pHLA) complexes, via their T cell receptors (TCRs), has been revealed using crystal structures. Indeed, X-ray crystallography, using the soluble version of both the TCR and pHLA, has shed light onto this intricate interaction essential to T cell activation, critical for host survivor. In this chapter, we describe the method to produce soluble TCRs.
