Three new alkaloids from Orychophragmus violaceus seeds were identified. Compound 3 showed significant ferroptosis inhibition by reducing reactive oxygen species and lipid peroxides, offering potential pharmacological applications.
Area of Science:
- Phytochemistry
- Pharmacology
- Cell Biology
Background:
- Orychophragmus violaceus seeds are a source of bioactive compounds.
- Ferroptosis is an iron-dependent cell death pathway implicated in various diseases.
- Understanding the phytochemistry and pharmacological activities of O. violaceus can lead to new therapeutic agents.
Purpose of the Study:
- To isolate and elucidate the structures of new and known alkaloids from O. violaceus seeds.
- To evaluate the ferroptosis inhibition activity of the isolated alkaloids.
- To investigate the underlying mechanisms of ferroptosis inhibition by active compounds.
Main Methods:
- Isolation of alkaloids using chromatographic techniques.
- Structure elucidation through spectroscopic data analysis (NMR, MS) and Electronic Circular Dichroism (ECD) calculations.
- In vitro assessment of ferroptosis inhibition using human intestinal epithelial cells (HIEC-6) and erastin induction.
- Measurement of reactive oxygen species (ROS), Fe2+, lipid peroxide (LPO), and glutathione (GSH) levels.
Main Results:
- Three new alkaloids (1-3) and three known alkaloids (4-6) were successfully isolated and characterized.
- Compound 3 demonstrated specific inhibition of erastin-induced ferroptosis in HIEC-6 cells.
- Compound 3 significantly reduced erastin-induced ROS, Fe2+, and LPO accumulation.
- Compound 3 promoted glutathione (GSH) production, counteracting ferroptosis.
Conclusions:
- The study successfully identified novel alkaloids from O. violaceus seeds with significant ferroptosis inhibitory effects.
- Compound 3 exhibits promising therapeutic potential for conditions associated with ferroptosis.
- These findings contribute to the understanding of O. violaceus phytochemistry and its pharmacological relevance.
