Abstract:
In this study, the relationship between extract condition, structural characteristics, spatial conformation and gel property of pectin was explored. A series of pectin (CPPs) was extracted at different pH (1.1-2.1). The structure, conformation and the gel properties of CPPs were analyzed by monosaccharide composition, small angle X-ray scattering (SAXS) and rheological analysis. With the increase of extract pH, degree of esterification (DE) increased from 39.02% to 68.52%, and the content of arabinose in Rhamnogalacturonan acid I (RG-I) domain increased from 2.51% to 15.12%. The different DE and RG-I side chain content led to the difference in molecular weight (Mw) (449.37-1266.74 kDa). Pectin with moderate DE and RG-I side chain had extended conformation. Extended conformation promoted the molecular interaction during gelation, which positively affect the gel property. This study revealed the connection between extraction condition, structural characteristics, spatial conformation and gel properties of pectin through various characterization methods, providing a new perspective for the extraction and application of pectin.
