Ecotoxicological impact and biological interactions of cathinone-derived psychoactive substances through quantitative

This study evaluates ecotoxicological impact and biological interactions of Cathinone (Cath), Methcathinone (MC), Amfebutamone (TBCP), α-Pyrrolidinovalerophenone (α-PVP), Methylone (MDMC), and Methylenedioxy pyrovalerone (MDPV) using QSAR and AOP-based predictive modelling. Cathinone has a higher solubility and atmospheric oxidation half-life, while MDPV exhibits maximum lipophilicity, environmental persistence (except in air), and bioconcentration. TBCP has the highest bioaccumulation potential. The BIOWIN model predicted that MDMC degrades rapidly under aerobic conditions, whereas all cathinone compounds showed slower degradation under anaerobic conditions. All compounds demonstrated the ability to cross the blood-brain barrier, while Cath, MC, MDMC, and MDPV have the potential to be carcinogenic. Additionally, MDPV is predicted to be immunogenic. Although all compounds exhibited poor dermal permeability, they demonstrated high intestinal absorption. The key interacting genes of six cathinone compounds were identified, and corresponding pathway mapping and molecular docking were done. Molecular docking revealed strong MDPV-protein interactions, suggesting significant biological and toxicological effects.
