Abstract:
Citrus maxima cv. Shating (pomelo) is one of the most popular pomelo varieties in Malaysia. We applied induced mutation breeding to broaden its genetic base, producing four Shating-derived mutant clones (2KRP6, 2KRP7, 2KRP9, and 3KRP4). While phenotypic information on Shating and these four mutants has been generated, genomic basis for this variety and its mutant lines remains limited. This scarcity of genomic data limits the identification of important genes and molecular markers, which is needed to facilitate future pomelo breeding programs. To address this issue, whole genome resequencing was conducted on Citrus maxima cv. Shating and its four mutant derivative clones (S2KRP6, S2KRP7, S2KRP9 and S3KRP4) to explore genome-wide variations induced by gamma irradiation. Despite the agricultural and commercial value of pomelo, particularly in Southeast Asia, comprehensive genomic information for this species and its mutant lines remains limited. In this study, genomic DNA from the parental cultivar and its mutants was sequenced using the Illumina NovaSeq X platform, generating high-quality paired-end reads with approximately 30× coverage per sample. The genome data enables the identification of single nucleotide polymorphisms (SNPs), insertions and deletions (InDels), and other structural variants that may be associated with phenotypic differences among the clones. These genome-wide variants offer valuable insights for mutation tracking, candidate gene discovery, and the development of molecular markers in pomelo improvement programs. All raw sequencing data in FASTQ format have been deposited in the European Nucleotide Archive (ENA) under the following PRJEB93835 accession numbers.
