Structural variation and rheological behaviour of pectin from Premna microphylla Turcz based on high-pressure

Abstract:
In this study, the effects of high-pressure homogenization treatment at 0-120 MPa on ammonium oxalate-extracted pectin from Premna microphylla Turcz were investigated. Increasing homogenization pressure reduced the weight-average molecular weight from 151.1 to 104.3 kDa and the degree of esterification from 8.49% to 5.60% while increasing the galacturonic acid content from 66.60% to 73.24%. The chain conformation transitioned from a semiflexible structure to irregular coils. The average particle size decreased from 886.9 to 173.6 nm, and the polydispersity index declined from 0.563 to 0.326. The modified pectin exhibited enhanced shear-thinning behavior, reduced thixotropy, and improved thermal stability. Antioxidant activities were significantly enhanced, with the IC50 values for 1,1-diphenyl-2-picryl-hydrazyl(DPPH)radical,2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS), and hydroxyl radicals decreasing from 3.58, 7.19, and 4.72 mg/mL to 2.61, 5.95, and 3.49 mg/mL, respectively. Overall, high-pressure homogenization effectively modified the structure and functionality of pectin, demonstrating its potential for food applications.
