Molluscan allergies pose increasing food safety concerns, yet reliable detection methods remain limited. This study developed a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method targeting tropomyosin to quantify molluscan allergens in foods. Using bioinformatics and liquid chromatography-high-resolution mass spectrometry (LC-HRMS), we identified two common peptides (LAITEVDLER, EDSYEETIR) and species-specific markers, evaluating their sensitivity, stability, and processing resistance. Matrix-matched calibration with isotope-labeled standards enhanced quantitative accuracy. Method validation demonstrated high specificity, with limits of detection (LOD) of 1.5-3 mg/kg, limits of quantification (LOQ) of 5-10 mg/kg in dumplings, excellent linearity (R2 > 0.995) from 5 or 10-500 mg/kg, recoveries of 70.4%-95.1%, and precision (RSD < 11.5%). Analysis of 25 commercial seafood dumplings revealed molluscan allergens in 68% of samples, with 82.4% of positive samples exceeding allergic reaction thresholds (ED05, 20 mg). This validated method provides a sensitive tool for molluscan allergen monitoring in complex food matrices.
