In this issue of Developmental Cell, Chovatiya et al. introduce PReCIS-seq, a method that captures RNA polymerase II dynamics with cell-type precision in intact tissue. Applying this method to mouse epidermis reveals a promoter-controlled pausing mechanism that differentially regulates genes governing lineage identity versus cellular safeguarding during development and adult homeostasis.
