Abstract:
Peimine (PM), a steroidal alkaloid predominantly found in Fritillaria species, has gained considerable attention due to its species-specific distribution and diverse pharmacological activities. Traditionally used for respiratory disorders, PM has demonstrated antitussive, anti-inflammatory, and anticancer effects through multi-target pathways. Major sources include F. thunbergii Miq. (FTB), F. ussuriensis Maxim. (FUB) and F. hupehensis (FHB), with its content varying across species, plant parts, and environmental conditions. Current evidence suggests that PM biosynthesis derives from cholesterol metabolism, with cytochrome P450 (CYP450) and glycosyltransferases involved in structural modification, but the full pathway remains unclear. Advances in genomics and enzyme characterization provide opportunities for synthetic biology-based production. However, low oral bioavailability and insufficiently defined pharmacokinetics limit its clinical potential. This review summarized recent progress on the distribution, biosynthesis, pharmacology, and application prospects of PM, aiming to support its rational development and utilization.
