Association study between IL-33 gene DNA methylation and Pediatric asthma

Objective:
Investigate IL-33 gene DNA methylation, its relationship with serum Th2 immune factors, and the rs4742170 SNP in childhood asthma, analyzing genetic, environmental, and epigenetic factors in asthma risk.
Methods:
MassARRAY assessed IL-33 DNA methylation and rs4742170 SNP; ELISA measured serum IL-4, IL-5, IL-13. Correlation analysis was conducted between DNA methylation, immune factors, and SNP, with a mediation effect of DNA methylation on asthma risk via environmental tobacco smoke (ETS) and SNP.
Results:
CONCLUSION: Hypomethylation at CpG_8 in IL-33 is linked to asthma risk. IL-33 regulates IL-4, IL-5, and IL-13 through different CpG sites. SNP rs4742170 and ETS may mediate asthma susceptibility via DNA methylation.
