Unbiased analyses of cancer genomes, transcriptomes, and proteomes have led to the identification of numerous genes that, when perturbed, contribute to the formation of cancer in humans. These unbiased analyses, by identifying drivers of cancer that localize to and function at the Golgi, have illuminated a previously unappreciated role for the Golgi in driving oncogenic phenotypes. Here we review some of the cancer drivers that act at the Golgi. We highlight the evidence that they are functionally important in cancer, and the evidence that they localize to and function at the Golgi. We also discuss functional interactions between these proteins, and review some of the proposed mechanisms by which they promote oncogenic phenotypes.
