Unexplained recurrent miscarriage (URM) is linked to cytotoxic Vδ2 γδT cells and vascular injury markers. Elevated Granzyme B⁺Perforin⁺ Vδ2 cells correlate with increased vascular damage, suggesting a novel immuno-vascular interaction in URM.
Area of Science:
- Immunology
- Reproductive Medicine
- Vascular Biology
Background:
- Unexplained recurrent miscarriage (URM) affects a significant portion of women, with underlying immune and vascular dysfunctions implicated.
- Peripheral blood Vδ2 γδT cells play a role in immune responses, but their specific contribution to URM pathogenesis remains unclear.
Purpose of the Study:
- To investigate the association between the cytotoxic profiles of peripheral blood Vδ2 γδT cells and vascular injury markers in women with unexplained recurrent miscarriage (URM).
Main Methods:
- Flow cytometry was used to analyze γδT cell subsets and their expression of perforin and granzyme B in 30 URM patients and 30 controls.
- Serum levels of vascular injury markers, including sFlt-1, sEng, vWF, LDH, and VCAM-1, were measured using ELISA.
Main Results:
- URM patients showed a significantly elevated frequency of Granzyme B⁺Perforin⁺ Vδ2 cells compared to controls.
- Serum levels of sFlt-1 and von Willebrand Factor (vWF) were significantly higher in the URM group.
- Positive correlations were observed between vascular injury markers (sFlt-1, vWF) and specific cytotoxic Vδ2 γδT cell populations.
Conclusions:
- Cytotoxic polarization of Vδ2 γδT cells is evident in URM and associated with vascular injury markers.
- These findings suggest a potential immuno-vascular interaction involving perforin-expressing Vδ2 cells in the pathogenesis of URM.
- This study offers hypothesis-generating insights for future research and potential therapeutic strategies in URM.
