Abstract:
Triple-negative breast cancer (TNBC) accounts for 15-20% of invasive breast cancers and represents a highly heterogeneous, aggressive subtype with poor prognosis and limited treatment options, necessitating the identification of novel therapeutic targets to improve clinical outcomes. Trophoblast cell-surface antigen 2 (Trop2), a calcium signal transducer, is frequently overexpressed in TNBC (approximately 88% of cases) while exhibiting minimal expression in normal tissues. Its overexpression is significantly associated with tumor invasion, metastasis, and unfavorable prognosis. Antibody-drug conjugates (ADCs) targeting Trop2 have demonstrated significant clinical potential. This article systematically reviews the clinical research progress of Trop2-targeted ADCs, aiming to provide evidence-based insights for improving the prognosis of TNBC patients.
Insights
Triple-negative breast cancer (TNBC) is aggressive with few treatments. Trop2-targeted antibody-drug conjugates (ADCs) show promise for improving outcomes in TNBC patients by targeting Trop2-overexpressing tumors.
Area of Science:
- Oncology
- Translational Medicine
- Drug Development
Background:
- Triple-negative breast cancer (TNBC) is an aggressive cancer subtype with limited therapeutic strategies.
- Trophoblast cell-surface antigen 2 (Trop2) is overexpressed in TNBC (88% of cases) and linked to poor prognosis.
- Trop2 overexpression correlates with tumor invasion and metastasis.
Purpose of the Study:
- To review the clinical progress of Trop2-targeted antibody-drug conjugates (ADCs) for TNBC.
- To provide evidence-based insights for enhancing TNBC patient outcomes.
Main Methods:
- Systematic review of clinical research on Trop2-targeted ADCs.
- Analysis of Trop2 expression in TNBC and normal tissues.
- Evaluation of Trop2-ADC clinical trial data.
Main Results:
- Trop2 is a promising therapeutic target due to its high expression in TNBC and low expression in normal tissues.
- Trop2-targeted ADCs have shown significant clinical potential in treating TNBC.
- ADC efficacy is linked to Trop2 expression levels and tumor characteristics.
Conclusions:
- Trop2-targeted ADCs represent a promising therapeutic avenue for TNBC.
- Further research and clinical trials are needed to optimize ADC therapy for TNBC.
- Targeting Trop2 may improve clinical outcomes for patients with this aggressive cancer subtype.
