Structure revision represents a cornerstone and an integral component of natural product chemistry. In this study, the planar structure of furanonaphthoquinone I (FNQI, 1) was revised, and its absolute configuration was determined through chemical derivatization, analysis of X-ray crystallography, and electronic circular dichroism (ECD) calculations. The revision of the chemical structure of 1 to 1a led to a corresponding reassessment of its biosynthetic pathway. Biological investigation revealed that 1a exhibited noncytotoxicity against human cancer cells and antibacterial activity against Gram-positive bacteria in vitro and in vivo.
