Novel XO inhibitory peptides from tiger shrimp (Penaeus monodon): Exploration, sequence optimization, inhibition type

Abstract:
Hyperuricemia (HUA) is gradually becoming one of the major problems threatening human health. Inhibition of xanthine oxidase (XO) activity represents an effective therapeutic strategy for managing HUA. In this study, four XO inhibitory peptides were obtained from tiger shrimp by using enzymatic hydrolysis and virtual screening. Among them, WENWTK exhibited the strongest XO inhibitory activity, with an IC50 value of 2.77 ± 0.06 mmol/L. The molecular mechanism of WENWTK was further investigated through molecular docking, molecular dynamics simulations (MD) and circular dichroism (CD) analysis. The inhibition kinetics of WENWTK was studied, revealing that it acts as a reversible mixed-type competitive inhibitor. Finally, rational design of sequence was performed through targeted substitution of residues in WENWTK with Trp. The optimized peptide, WEWTW, exhibited an improved IC50 value of 1.73 ± 0.13 mmol/L. This study underscores the potential of tiger shrimp peptides and a combined computational-rational design approach for developing anti-hyperuricaemic agents.
