Objectives:
This study investigated the role of miR-146a-5p rs2910164 polymorphism in attention deficit hyperactivity disorder (ADHD).
Methods:
This study included 180 ADHD children and 122 healthy individuals aged 6-12 years. The ADHD children were hospitalized for treatment while healthy individuals were recruited from a local primary school. ADHD patients received 24-week MPH administration, and treatment response was evaluated via ADHD-RS and CGI-S or CGI-I. miR-146a-5p levels were quantified by qRT-PCR. Multivariate logistic regression was employed for the independent risk factors including rs2910164 polymorphism and gender. MiR-146a-5p rs2910164 was genotyped via TaqMan Genotyping method.
Results:
In ADHD children, GG carriers exhibited markedly higher miR-146a-5p levels than GC/CC carriers. A similar genotype-related elevation was also observed but was less pronounced in control. The effect allele G and male subgroup were significantly associated with ADHD susceptibility. After 24-week MPH administration, GC and CC carriers showed higher odds of successful treatment than GG individuals in both ADHD-RS and CGI-I assessment. miR-146a-5p was increased in ADHD children and showed diagnostic promise.
Conclusion:
The rs2910164 polymorphism was related to ADHD children in the Chinese population. However, further validation in independent and larger populations is necessary to confirm this relationship and establish its generalizability.
