Abstract:
We report the cytotoxic effects of Lansbermin-I, a disintegrin from Porthidium lansbergii lansbergii venom, on HeLa cervical cancer cells but not on leukemic, non-tumorigenic peripheral blood mononuclear cells (PBMCs) or fibroblast lines. Lansbermin-I reduced HeLa cell viability in a concentration-dependent manner, impaired adhesion to fibronectin and collagen, and inhibited migration and clonogenicity. Docking analysis suggested interaction with the RGD domain of integrin α5β1. These results identify Lansbermin-I as a potential antitumor prototype against cervical cancer.

Integrins
Some ECM proteins assemble into a basement membrane to which the remaining components adhere. Proteoglycans typically form the bulk of the ECM while fibrous proteins, like collagen,...
Activation of Integrins
In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding...
