Real-time reverse transcription-polymerase chain reaction, or Real-time RT-PCR, is an analytical tool used to determine the expression level of target genes. Summary

Fungal translocation occurs in patients with alcohol use disorder (AUD), with Candida albicans frequently detected in blood. Alcohol withdrawal did not significantly alter fungal DNA levels, suggesting translocation may be independent of gut barrier integrity.

Background:

  • Previous translocation studies primarily focused on bacteria, overlooking fungal roles.
  • Fungal translocation across the gut barrier is a potential contributor to various diseases.

Purpose of the Study:

  • To investigate fungal translocation in patients with alcohol use disorder (AUD).
  • To determine if alcohol withdrawal impacts fungal translocation.
  • To explore the relationship between fungal translocation and intestinal integrity markers.

Main Methods:

  • Sixty-five AUD patients provided blood samples at baseline and 3, 6 weeks post-withdrawal.
  • Fungal DNA was detected using qRT-PCR with panfungal primers and subsequently sequenced.
  • Intestinal integrity markers were assessed for correlation with fungal translocation.

Main Results:

  • Fungal DNA was detected in 30.9% of tested AUD patients, predominantly Candida albicans.
  • No significant changes in fungal DNA levels were observed during alcohol withdrawal.
  • No correlation was found between intestinal integrity markers and fungal translocation.

Conclusions:

  • Fungal elements, particularly Candida albicans, can translocate into the bloodstream of AUD patients.
  • Alcohol withdrawal does not appear to significantly affect fungal translocation in this cohort.
  • Fungal translocation may occur independently of compromised gut barrier integrity in AUD.
