Abstract:
Sepsis-induced acute kidney injury (S-AKI) is a common, life-threatening clinical complication with complex, incompletely understood pathophysiological mechanisms. Historically, the three main types of regulated cell death (RCD) were necroptosis, apoptosis, and pyroptosis. Recent discoveries of significant molecular overlap between these pathways have suggested that PANoptosis represents a combined inflammatory RCD mechanism. Accumulating evidence suggests that PANoptosis contributes to the progression of S-AKI. To clarify underlying mechanisms and guide innovative therapeutic methods, this review summarizes recent findings on the pathogenesis of S-AKI, with an emphasis on the growing significance of PANoptosis'.
Insights
Sepsis-induced acute kidney injury (S-AKI) involves complex cell death pathways. This review highlights PANoptosis, a combined inflammatory cell death, as a key contributor to S-AKI progression and potential therapeutic target.
Area of Science:
- Nephrology and Immunology
- Molecular Biology and Cell Death Mechanisms
Background:
- Sepsis-induced acute kidney injury (S-AKI) is a critical clinical condition with poorly understood mechanisms.
- Traditional regulated cell death (RCD) types include necroptosis, apoptosis, and pyroptosis.
- PANoptosis emerges as a unified inflammatory RCD pathway due to molecular overlaps between necroptosis, apoptosis, and pyroptosis.
Purpose of the Study:
- To review and synthesize recent findings on S-AKI pathogenesis.
- To emphasize the emerging role of PANoptosis in S-AKI progression.
- To provide insights for developing novel therapeutic strategies against S-AKI.
Main Methods:
- Literature review of recent research on Sepsis-induced acute kidney injury.
- Analysis of molecular mechanisms underlying regulated cell death pathways.
- Focus on the integration of necroptosis, apoptosis, and pyroptosis into PANoptosis.
Main Results:
- PANoptosis is increasingly recognized as a significant contributor to S-AKI.
- The complex interplay of RCD pathways is central to S-AKI pathophysiology.
- Understanding PANoptosis offers new avenues for S-AKI treatment.
Conclusions:
- PANoptosis represents a critical, unified inflammatory cell death mechanism in S-AKI.
- Further research into PANoptosis is essential for advancing S-AKI treatment.
- Targeting PANoptosis may offer innovative therapeutic approaches for sepsis-related kidney injury.
