Preclinical Development: Overview

6.4K
Preclinical development consists of a series of tests that ensure the safety and efficacy of a new therapeutic compound before it is tested in humans. There are four main phases to this process. First, safety pharmacology tests are conducted to ensure the drug does not produce any acutely harmful effects. These tests examine parameters such as bronchoconstriction, cardiac dysrhythmias, blood pressure changes, and ataxia. Next, preliminary toxicological testing is performed to determine the...

Clinically Relevant Drug Product Specifications: Methods of Establishment

283
Product specifications define the acceptable quality of a pharmaceutical product by ensuring identity, purity, potency, and strength. These specifications serve as benchmarks during development, manufacturing, and post-approval quality control. Clinically relevant specifications are particularly important because they directly relate to a drug's safety and efficacy in clinical use.Dissolution studies are critical biopharmaceutic tools that link in vitro behavior to in vivo performance. They...

Area of Science:

  • Cosmetic Science
  • Dermatology
  • Sustainable Materials

Background:

  • Insect oils are gaining traction as sustainable cosmetic ingredients due to their rich bioactive lipid content.
  • Cricket oils offer a potential alternative to traditional cosmetic oils.
  • This study investigates the potential of cricket oils in cosmetic applications, specifically sunscreens.

Purpose of the Study:

  • To characterize oils from three cricket species.
  • To evaluate the safety, biological activities (antioxidant, anti-inflammatory), and performance of cricket oils in sunscreen formulations.
  • To determine the efficacy of cricket oil-based sunscreens in providing UV and blue light protection.

Main Methods:

  • Oils were extracted from Gryllus bimaculatus, Teleogryllus mitratus, and Acheta domesticus.
  • Fatty acid composition was analyzed using gas chromatography-mass spectrometry.
  • Safety was assessed via cytotoxicity and hen's egg chorioallantoic membrane assays.
  • Antioxidant and anti-inflammatory activities were evaluated using reactive oxygen species and nitric oxide assays.
  • Teleogryllus mitratus oil (TMO) was incorporated into sunscreen formulations with UV filters.

Main Results:

  • All cricket oils were non-cytotoxic and non-irritating.
  • TMO demonstrated potent antioxidant and anti-inflammatory properties.
  • TMO provided measurable UVA protection (PPD/UVA-PF = 12.1, PA+++).
  • Sunscreen formulations with TMO showed enhanced Sun Protection Factor (SPF) up to 40.51 and PPD/UVA-PF up to 39.17 compared to olive oil controls.
  • Inclusion of pigments further boosted SPF and blue light protection.

Conclusions:

  • Teleogryllus mitratus oil (TMO) is a safe and effective multifunctional ingredient for cosmetic formulations.
  • TMO significantly enhances the photoprotective performance of sunscreens.
  • The use of TMO supports the development of sustainable cosmetic products.
