Cell Signaling in Plants01:25

Cell Signaling in Plants

7.0K
Plant cells communicate to coordinate their cycle of growth, flowering and fruiting, and activities in roots, shoots, and leaves in response to the changing environmental conditions. Plant signaling is distinct from animal signaling. Plants primarily utilize enzyme-linked receptors, whereas the largest class of cell-surface receptors in animals are G-protein coupled receptors (GPCRs). Unlike animals, receptor tyrosine kinases are rare in plants. Instead, plants have a diverse class of...

Receptor Tyrosine Kinases01:26

Receptor Tyrosine Kinases

20.7K
Receptor tyrosine kinases or RTKs are membrane-bound receptors that phosphorylate specific tyrosine on protein substrates. RTKs regulate cellular growth, differentiation, survival, and migration. They contain an extracellular ligand binding domain, a transmembrane domain, and a cytosolic tail with intrinsic kinase activity. Several extracellular signaling molecules activate RTKs in one or more ways and relay the signal downstream. Ligands such as platelet-derived growth factor (PDGF) or...

Rab Proteins01:14

Rab Proteins

5.4K
Rab proteins constitute the largest family of monomeric GTPases, of which 70 members are present in humans. Rab proteins and their effectors regulate consecutive stages of vesicle transport such as vesicle transport, docking, and fusion to the correct recipient membrane.
Rab proteins switch between a cytosolic, GDP-bound inactive state and a membrane-anchored, GTP-bound active state. By themselves, Rabs show slow rates of GDP/GTP exchange and GTP hydrolysis. Thus, Rab proteins are considered...

PI3K/mTOR/AKT Signaling Pathway01:22

PI3K/mTOR/AKT Signaling Pathway

6.3K
The mammalian target of rapamycin  (mTOR) is a serine/threonine kinase that regulates growth, proliferation, and cell survival in response to hormones, growth factors, or nutrient availability. This kinase exists in two structurally and functionally distinct forms: mTOR complex 1  (mTORC1) and mTOR complex 2  (mTORC2). The first form (mTORC1) is composed of a rapamycin-sensitive Raptor and proline-rich Akt substrate, PRAS40. In contrast,  mTORC2 consists of a...

Summary

Potato receptor kinase RDA2 proteins confer resistance against the pathogen Phytophthora infestans. This plant immunity is suppressed by pathogen RXLR effectors, highlighting a key battle in plant defense.

Area of Science:

  • Plant immunity
  • Molecular plant-pathology
  • Plant-microbe interactions

Background:

  • Plants use pattern recognition receptors (PRRs) to detect pathogen-associated molecular patterns (PAMPs), initiating pattern-triggered immunity (PTI).
  • Late blight, caused by Phytophthora infestans, severely impacts potato crops globally.
  • The PAMP P. infestans cell wall ceramide D induces reactive oxygen species (ROS) and is recognized by Arabidopsis RDA2.

Purpose of the Study:

  • To identify and characterize potato RDA2 homologs and their role in plant defense against P. infestans.
  • To investigate the mechanism by which P. infestans RXLR effectors interfere with potato immunity.

Main Methods:

  • Sequence alignment to identify potato RDA2 genes.
  • Gene expression analysis in potato plants infected with P. infestans.
  • Transient expression assays in Nicotiana benthamiana to assess StRDA2A function and effector interactions.

Main Results:

  • Potato RDA2 genes were identified, and their expression varied upon P. infestans infection.
  • StRDA2A expression induced cell death and enhanced disease resistance in N. benthamiana, unlike its kinase-dead mutant.
  • P. infestans RXLR effectors suppressed StRDA2A-mediated cell death.

Conclusions:

  • Potato RDA2 receptor kinases play a crucial role in conferring resistance against P. infestans.
  • P. infestans RXLR effectors target and inhibit StRDA2A function to facilitate infection.
  • Understanding this interaction is vital for developing strategies to enhance potato disease resistance.
