Purpose of the Study:

  • To compare the efficacy of cryopreserved human amniotic membrane (cHAM) versus Biodentine for early pulp biomineralization.
  • To evaluate the biomineralization potential of cHAM and Biodentine using a tooth culture model.

Main Methods:

  • A tooth culture model was established and validated for pulp cell viability.
  • Pulp capping was performed on extracted teeth using cHAM or Biodentine.
  • Biomineralization was assessed at 14 and 28 days using micro-computed tomography (micro-CT) and histological analysis.

Main Results:

  • cHAM resulted in significantly thicker and larger volumes of mineralization (720 μm, 0.7 mm³) compared to Biodentine (630 μm, 0.46 mm³) at 28 days.
  • Histological analysis showed homogeneous and continuous dentine formation with cHAM, versus discontinuous mineralization with Biodentine.
  • Pulp cell viability in the culture model was 66.1% at 28 days and 38.4% at 50 days.

Conclusions:

  • Cryopreserved human amniotic membrane (cHAM) demonstrates superior early pulp biomineralization potential compared to Biodentine.
  • cHAM may represent a promising alternative for pulp capping procedures requiring enhanced biomineralization.
  • Further research is warranted to confirm these findings and explore clinical applications.
