Methodological validation of microbial limit tests for high-viscosity and common pharmaceutical excipients

Microbial contamination of pharmaceutical excipients remains a critical precursor to finished product failure, which necessitates rigorous microbial limit testing of pharmaceutical excipients. However, high-viscosity excipients present distinct analytical challenges, including hygroscopicity, swelling upon hydration, and agglomeration. These physicochemical properties often impede sample homogenization, obstruct membrane filtration, and compromise microbial recovery. To address these limitations, this study established and validated robust microbial limit testing protocols for nine specific excipients-four characterized by high viscosity and five commonly utilized in regional manufacturing-strictly adhering to the Chinese Pharmacopoeia (ChP) General Principles. Method suitability was demonstrated through independent replicate testing, yielding recovery rates between 0.5 and 2.0 for all samples. Microbial enumeration validation encompassed Total Aerobic Microbial Count (TAMC) using Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus subtilis, Candida albicans, and Aspergillus niger, and Total Yeast and Mold Count (TYMC) using C. albicans and A. niger. Furthermore, methods for specified microorganisms (Escherichia coli and Salmonella) have been validated, confirming that the test bacteria grow well and that the method is effective and feasible. Beyond methodological validation, this work bridges microbial limit testing gaps in current compendial standards for these specific excipients. By integrating these validated protocols with a comprehensive assessment of microbial control practices within Yunnan Province's pharmaceutical sector, this study offers a practical method to resolve persistent technical bottlenecks and supports enhanced regulatory oversight and quality assurance strategies.
