Further phytochemical investigation on Rhodomyrtus tomentosa leaves led to the isolation of three new acylphloroglucinol trimers (APTs), named rhodotomentotrimers G-I (1-3). Their structures and absolute configurations were unambiguously established by means of comprehensive spectroscopic analysis (NMR and HRMS), NMR calculations and DP4+ probability analyses, ECD simulations, and single-crystal X-ray diffraction. All these isolates displayed antibacterial effects on three methicillin-resistant Staphylococcus aureus (MRSA) strains and vancomycin-resistant Enterococci (VRE) with an MIC value of 0.78 μg/mL, respectively. Mechanism investigations indicated that these isolates might exert bacteriostatic and bactericidal effects by disrupting cell membranes. These findings could greatly enrich the chemistry diversity of phloroglucinol components and provide potential candidates for the development of antibacterial agents.
