Chimeric antigen receptor (CAR) T cell therapy targeting B cell maturation antigen (BCMA) shows promise for treating refractory systemic lupus erythematosus (SLE). This novel approach demonstrated rapid clinical improvement and tolerability in a small patient cohort.
Area of Science:
- Immunology
- Cell Therapy
- Autoimmune Diseases
Background:
- Systemic lupus erythematosus (SLE) is a complex autoimmune disorder with limited treatment options for refractory cases.
- Characterized by autoantibodies and complement activation, SLE follows a chronic relapsing-remitting course.
Purpose of the Study:
- To evaluate the feasibility, tolerability, and short-term efficacy of B cell maturation antigen (BCMA) chimeric antigen receptor (CAR) T cell therapy in patients with refractory SLE.
Main Methods:
- Three female patients with refractory SLE received autologous T cells engineered with a lentiviral anti-BCMA CAR vector.
- CAR T cells were administered intravenously at a dose of 40 million cells on days 0 and 2.
Main Results:
- All patients experienced significant clinical improvement by day 13.
- Normalization of laboratory parameters was observed in all participants.
- The BCMA CAR-T cell therapy was found to be feasible and well-tolerated.
Conclusions:
- BCMA CAR-T cell therapy presents a potential new treatment strategy for systemic lupus erythematosus.
- The therapy demonstrated short-term efficacy and tolerability, warranting further clinical investigation.
- This approach offers hope for patients with treatment-resistant SLE.
