Trihybrid Crosses
Some of Mendel’s crosses examined three pairs of contrasting characteristics. Such a cross is called a trihybrid cross. A trihybrid cross is a combination of three individual monohybrid crosses. For example, plant height (tall vs. short), seed shape (round vs. wrinkled), and seed color (yellow vs. green).
The F1 generation plants of a trihybrid cross are heterozygous for all three traits and produce eight gametes. Upon self-fertilization, these gametes have an equal...

Crop cultivation has a long history in human civilization, with records showing the cultivation of cereal plants beginning at around 8000 BC. This early plant breeding was developed primarily to provide a steady supply of food.

Area of Science:

  • Plant genetics
  • Crop science
  • Molecular biology

Background:

  • Golden Promise (GP) is a key barley reference genotype for transformation but has a long generation cycle and stress susceptibility due to a Photoperiod-H1 (Ppd-H1) mutation.
  • A previous introgression line, GP-fast, showed improved flowering time and stress resilience by introducing a wild-type Ppd-H1 allele.

Purpose of the Study:

  • To develop an isogenic, fast-cycling barley genotype (GP-rapid) with retained regeneration and transformation capabilities.
  • To reduce the generation time of the Golden Promise barley cultivar for enhanced research applications.

Main Methods:

  • Generated GP-rapid through two backcrosses of GP-fast to the GP background, isolating a single introgression at the Ppd-H1 locus.
  • Evaluated generation time under speed breeding conditions.
  • Assessed transformation efficiency using CRISPR/Cas9-mediated genome editing.

Main Results:

  • GP-rapid achieved a generation time of 63 days, a 25% reduction compared to GP's 84 days.
  • The new genotype retained amenability to Agrobacterium-mediated transformation.
  • The final introgression at the Ppd-H1 locus was approximately 0.6 Mbp on chromosome 2H.

Conclusions:

  • GP-rapid is a valuable, fast-cycling isogenic line for barley research.
  • This development facilitates the efficient generation of transgenic and gene-edited barley plants.
  • The optimized genotype accelerates research timelines in barley molecular breeding.
